I study plant proteins which inhibit hydrolytic enzymes of various organisms:
Inhibitors are of interest as:
The inhibitors are well studied in various plants, but, generally, molecular structures of single inhibitors are studied much more then their natural variability in plant taxa.
The simple and sensitive methods for detection of inhibitors of alpha-amylases, serine and cysteine proteinases and lectins among plant proteins separated by electrophoresis, isoelectric focusing or thin layer gel-filtration were elaborated (Konarev, 1985, 1986; Konarev et al., 1991, 2000, 2002, Konarev and Lovegrove, 2012). These methods were used in study of the polymorphism and natural variability of main inhibitor systems in wild and cultivated species of wheat and related cereals, rice, potato, Vigna and Phaseolus, sunflower, various Compositae, Apiaceae, other Asteridae and for identification of novel inhibitor types e.g. small (1.5 kDa) cyclic trypsin inhibitor from sunflower seeds, low-molecular trypsin Inhibitor with helix-turn-helix protease Inhibitory motif from seeds of
Veronica (VhTI) or serine proteinase inhibitor from seeds of ancient gymnosperm plant, Cycas siamensis.
Detection methods were developed for proteinases as well that made it easier the identification and purification of a novel glutenin-specific salivary gland proteinase of Sunn pest (Eurygaster integriceps Put.) responsible for wheat gluten degradation and bread quality deterioration (Konarev et al., 2011, Konarev and Lovegrove, 2012).

| Polymorphism of insect alpha-amylase inhibitors in diploid wheats collected in Armenia and Azerbaidjan.

B, thin layer gel filtration. Inhibitors of Tenebrio molitor amylase were detected with use of PAAG/starch/amylase replicas
|Variability of trypsin inhibitors in Vigna and Phaseolus species.
Isoelectric focusing of seed proteins followed by detection of inhibitors with use of gelatin replicas method.
|The polymorphism of chymotrypsin/subtilisin inhibitors in several bread wheat varieties. Endosperm proteins were extracted with water and separated by isoelectric focusing in PAG. Protein bands were transferred from gel to the gelatine replica by diffusion and the latter was developed by chymotrypsin.|
|More than 500 species of Compositae and other Asteridae were screened for proteinase inhibitors. Cyclic low-molecular trypsin inhibitor consisting only of 14 amino acid residues was found only in seeds of Helianthus and Tithonia species. It appeared to be the smallest and most potent trypsin inhibitor ever found in plants.|
